چکیده

One of the main reasons for the high drug attrition rate in oncology is the poor clinical predictive power of 2D cancer cell lines cultures in vitro which are the standard assay used as screening assay to select new chemical entities (NKE) with potent anticancer activity. Therefore, there is increasing interest in developing 3D in vitro cell culture systems, as a primary screening assays which would represent a biologically more relevant assay system, due to similarity to physiological conditions in which tumors growth in living organism. Very important is to develop reproducible 3D cell line culture in vitro assays, feasible for the primary screening of NKEs platforms. Within this manuscript we have tested small platform of selected benzo[b]thieno, thieno[2,3-b]thiophene, and thieno[3,2-b]thiophene 2-carboxanilides, as well as their cyclic derivatives [2,3-c]quinolones, which already showed anti-proliferative activity on other cancer cell lines in 2D system. Platform was tested in 2D and 3D cancer cell culture assays on three human breast cancer cell lines (SK-BR-3, MDA-MB-231, T-47D). Those cell lines were selected on the basis of ability to growth and form cell spheres and also on different sensitivity to chemotherapeutic agents. We used doxorubicin as control compound due similar mode of action, (specific intercalation with the DNA double helix), as tested compound probably have. Obtained results in some cases showed significant disagreement, indicating that in early screening selection of active compounds only on 2D activity basis we could pick up false positive compounds (active only on 2D cell culture lines and not on 3D cell lines for which it is clamed that are more similar to real physiological conditions for tumor growth). One possibility for obtained discrepancy of results obtained on 2D and 3D cell cultures could be physico-chemical properties of compounds. Therefore, we analyzed some physico-chemical properties of active compounds as chrom logD values and calculated structural parameters: number of hydrogen bond donors and acceptors, calculated logP and logD values, molecular weight, ionization constants (pKa), number of aromatic rings, number of rotatable bonds and polar surface area. Association of physico-chemical descriptors with observed anti-proliferative activity has been investigated. Basicity, molecular weight and number of H-bond donors are found to be main factors contributing to the anti-proliferative effect of investigated compounds for both 2D and 3D cell cultures.

O-16: Comparison of Pre-Antral Follicle Culture Development during 2 Dimensional and 3 Dimensional Culture Systems

Background: Setting up an in vitro follicle culture system that resembles in vivo ovary condition has high value in research. Additionally, expression evaluation of folliculogenesis involved genes could lead us to the designing of better culture system. Materials and Methods: ovaries of 12-day-old female NMRI mice were removed, 100-130 μm pre-antral follicles were mechanically isolated from fre...

Comparison of the Ex Vivo Expansion of UCB-Derived CD34+ in 3D DBM/MBA Scaffolds with USSC as a Feeder Layer

Objective(s): Ex vivo expansion of hematopoitic stem cells is an alternative way to increase umbilical cord blood (UCB)-CD34+ cells for bone marrow transplantation. For this purpose demineralized bone matrix (DBM) and mineralized bone allograft (MBA) as two scaffolds based on bone matrix and stem cell niche, were simultaneously used to enhance the effect of human mesenchymal pro...

Induced Chondrogenic Differentiation of hESCs by hESC-Derived MSCs Conditioned Medium and Sequential 3D-2D Culture System

Background and Aims: It has been proven that human mesenchymal stem cells (MSCs) conditioned medium (hMSCs-CM) can influence human embryonic stem cells (hESCs) chondrogenic differentiation. In this study, we hypothesized that conditioned medium (CM) from hESCs-derived MSCs in a sequential 3D-2D culture system could facilitate the induction of chondrogenesis in hESCs. Materials and Methods: CM ...

Comparison of the Expression of Hepatic Genes by Human Wharton's Jelly Mesenchymal Stem Cells Cultured in 2D and 3D Collagen Culture Systems

Background: Human Wharton’s jelly mesenchymal stem cells (HWJMSCs) express liver-specific markers such as albumin, alpha-fetoprotein, cytokeratin-19, cytokeratin-18, and glucose-6-phosphatase. Therefore, they can be considered as a good source for cell replacement therapy for liver diseases. This study aimed to evaluate the effects of various culture systems on the hepatocyte-specific gene expr...

Wharton’s Jelly Mesenchymal Stem Cell: Various Protocols for Isolation and Differentiation of Hepatocyte-Like Cells; Narrative Review

There are several differentiation methods for mesenchymal stem cells (MSCs) into hepatocyte-like cell. Investigators reported various hepatic differentiation protocols such as modifying culturing conditions or using various growth factors/cytokines. In this literature review, we compared different MSCs extraction and isolation protocols from Wharton’s jelly (WJ) and explored various MSCs differ...
